Orangutans harbor ancient primate Alu

ScienceDaily (Apr. 29, 2012) ? Alu elements infiltrated the ancestral primate genome about 65 million years ago. Once gained an Alu element is rarely lost so comparison of Alu between species can be used to map primate evolution and diversity. New research published in BioMed Central's open access journal Mobile DNA has found a single Alu, which appears to be an ancestral great ape Alu, that has uniquely multiplied within the orangutan genome.

Analysis of DNA sequences has found over a million Alu elements within each primate genome, many of which are species specific: 5,000 are unique to humans, while 2,300 others are exclusive to chimpanzees. In contrast the orangutan lineage (Sumatran and Bornean orangutans) only has 250 specific Alu.

These tiny pieces of mobile DNA are able to copy themselves using a method similar to retroviruses. But, because this is an inexact process, a segment of 'host' DNA is duplicated at the Alu insertion sites and these footprints, known as 'target site duplications', can be used to 'identify' Alu insertions. Alu elements can be thought of as molecular fossils, and a shared Alu element sequence and location within the genome indicates a common ancestor.

Researchers from Louisiana State University, in collaboration with the Zoological Society of San Diego and the Institute of Systems Biology in Seattle, found a single Alu which is present in great apes, but absent from gibbons and siamang, and so was likely acquired after ancestors of these species diverged. This founder Alu element was found in an intron (non-coding DNA) on chromosome 7.

Subsequent copying of this Alu, visible as insertions into chromosomes 4, 17, and 12, are unique to orangutans, suggesting that the founder Alu inserted before orangutans separated from other members of the Hominidae (humans, gorillas, and chimpanzees). The Sumatran orangutan also has a copy of this Alu in chromosome 13, and has gained a daughter Alu, which rapidly expanded into chromosomes 21, 2b, and 17. These extra insertions are able to pinpoint the divergence of Sumatran and Bornean orangutans.

The ancestral Alu has been much less active in other great apes, but can still provide information about speciation. While still on chromosome 7 the Alu gained three mutations which can be traced to before the split of gorillas with humans and chimpanzees. It subsequently copied itself into chromosome 3 of humans, indicating that this must have occurred after humans split from bonobos and chimpanzees.

Prof Batzer, who led this research along with Jerilyn Walker and Miriam Konkel, explained "Despite otherwise low activity of Alu retrotransposition in orangutans, this ancestral Alu, still present on chromosome 7, has duplicated more rapidly in orangutans than other Hominidae and likely served as an ancient backseat driver that contributed to the recent orangutan-specific expansion of the Alu family."

Colts make Harnish final choice of 2012 draft

(AP) ? Mr. Irrelevant suddenly has a whole new meaning for Chandler Harnish.

The Northern Illinois quarterback who was taken with the 253rd and final pick of the draft Saturday didn't just get a title and a vacation. He got a chance to play for his favorite NFL team and potentially to be a backup for Andrew Luck.

First, he'll make the trip to Disneyland, appear in the Irrelevant Week parade in Newport Beach, Calif., and put his Colts license plate back on his car. Hey, it's OK -- he's an Indiana native.

"I'm happy to be wanted by a team and I'm happy to be an Indianapolis Colt because that's been my favorite team all the way through," he told local reporters on a conference call shortly after the selection was announced.

For the Colts and the NFL, it was a historic first.

Indianapolis opened the draft by taking Luck, the Stanford quarterback, with the No. 1 overall selection and ended the draft by taking another quarterback in Harnish. It was the first time since 1967 that a non-expansion team had the first and last picks in the draft and only the third time in league history.

Neither of the previous teams, Houston in 2002 and Philadelphia in 1949, chose a quarterback with both picks.

Paul Salada, who founded Irrelevant Week in 1976, made the announcement and held up a blue-and-white Colts jersey with No. 253 on it.

One thing is already certain: Unlike Luck, who was handed a No. 12 jersey with his name stitched on the back, Harnish will not wear No. 253 when he reports to next weekend's rookie mini-camp.

And for a guy who grew up playing at Norwell High School in northeastern Indiana, this was no joke, either.

"It's complete elation. It's surreal," Harnish said. "I'm so happy I'm a Colt. I'm really, just speechless."

Harnish set 30 school records at Northern Illinois, was a two-time all-Mid-American Conference selection, a three-time academic all-league choice and was chosen as the league's MVP. He led the Huskies to the league title and their fourth straight bowl game in 2011.

He finished his career with 8,944 yards passing, completed 61.9 percent of his passes with 68 touchdowns and 26 interceptions. He also ran for 2,238 yards and 24 TDs and his 11,927 yards in total offense rank third all-time in league history.

That was enough to persuade the Colts to take him, even at draft's end.

"It is relevant because otherwise you wouldn't burn a pick on somebody," new general manager Ryan Grigson said. "This person we took had tremendous success in the Mid-American Conference. He's got ability, and he was higher on our board than where we took him."

Plus, the Colts need him.

Until Thursday night, the only quarterbacks on Indy's roster were career backup Drew Stanton, who was acquired in an offseason trade with the New York Jets, and Trevor Vittatoe, with one year of pro experience. Pro Bowl receiver Reggie Wayne couldn't even remember his name and simply chose to call him "V."

The selection of Luck altered the depth chart, and the selection of Harnish gives the Colts another young quarterback to groom as a backup to Luck.

Harnish, who is 6-foot-2 and 220 pounds, couldn't have asked for a better situation.

"I was right there at the end, still hoping for the best and preparing for the worst," he said. "My agent and I were thinking we'd be looking into free agency.

"It looks like I have a little vacation and they tell me I have a parade," he added. "I don't know much else about what happens, but I'm excited to find out."

Harnish and the Colts weren't the only winner.

Team owner Jim Irsay also ran a contest on Twitter, offering $5,000 to the person who chose the name of the last of Indy's 10 selections this weekend. Shortly after the draft, Irsay tweeted that the winner was Billy Cardwell from Kokomo, Ind.

Neil Heywood: new details emerge of British business dealings of Chinese woman suspected of murder

Gu Kailai, who is in custody accused of poisoning Mr Heywood, struck a bizarre deal to import hot air balloons from Dorset to China ? and is accused of trying to illegally bring into Britain ?200,000 to pay for her son's public school fees.

The deal shows how Mrs Gu, whose husband Bo Xilai was until last month one of China's most powerful politicians, and Mr Heywood enjoyed a business relationship going back almost 15 years, suggesting he was deeply involved in her financial affairs. It has been claimed Mr Heywood may have been killed because he knew too much.

* Scotland Yard is set to conduct its own investigation into Mr Heywood's death on the orders of a London coroner. The Sunday Telegraph has learned that Dr Andrew Harris, coroner for south London, is considering writing to Ken Clarke, the Justice Secretary, requesting permission to hold an inquest under a special section of the Coroners Act.

* The Duke of York invited the Chinese ambassador to Buckingham Palace last week for private talks. A spokesman refused yesterday to disclose details of Prince Andrew's meeting with Liu Xiaoming, the Chinese ambassador, but its timing will raise speculation over concern over the death.

Only last May, the Duke met Bo Xilai in China in his capacity as Britain's special trade envoy.

* dramatic details emerged of blind dissident Chen Guangcheng's daring escape from house arrest to the US embassy in Beijing. Hu Jia, a friend of Mr Chen, told the Sunday telegraph how the dissident managed to evade capture in a police chase. Hours later, Mr Hu was himself detained by Chinese authorities

* China and the US are embroiled in a growing diplomatic row ahead of a visit this week by US Secretary of State Hillary Clinton. The Chinese government are putting pressure on Mrs Clinton to release Mr Chen from US protection but such a move would be severely criticised in Washington.

China's ruling elite has been shaken by both Mr Chen's escape and the investigations into Mr Bo and his wife, who are both accused of being at the centre of a network of multi million pound corruption.

Can you charge a Galaxy Tab 2 7" with a charger from Galaxy Tab 10.1?

In my opinion there is no harm to your device if the charger produce the enough voltage to charge the device. You have to keep in mind that the charger should not be low power producer as your device needed otherwise it will be harmful for that, but if the charger produce the same power or higher then it will be fine for device because if the charger produces high output voltage your device will only take how much it need.

New York Theatre Review: Carissa Cordes on Your Boyfriend May ...

Your Boyfriend May Be Imaginary is a new play by Larry Kurnofsky. The story opens on a party and follows thirty-something Marci (Darcy Fowler), who is trying to find her boyfriend in her pajamas through various connected people at various grown-up high school friend?s parties in the city. Marci?s quest is constantly hampered by her friends believing Marci is sometimes ?a little sad and a little strange? and her boyfriend is imaginary. Your Boyfriend features a whole host of characters; such as Cassandra (Risa Sarachan), her best friend (who?s not really), Beth (Kirsten Hopkins), who insists on using the formal Marcia and doesn?t hide her disdain, her gay roommate Toddwhatshisname (Debargo Sanyal), Carl (Zach Evenson) the only straight man at party in Chelsea, HunkyDave (Quinlan Corbett) who Marci was allegedly stalking, the divorce party of Paul and Paula Paul (Jordan Mahone and Danielle Slavick), Denise (Maya Lawson) who is engaged and seemingly obsessed with Marci, and last but not least, the not physically present boyfriend, Phillip Abramovich, an activist who lives off the grid.? Marci, who doesn?t own a cell phone and insists on being invisible or ?not really here?, is unsuccessful in her boyfriend finding mission, yet, she comes home with 3 cell phones, a tiara and lamp and she has turned all the characters on their side with honest observation. In the end it is Denise who calls Marci and shares a very personal story in camaraderie and the play ends in the exact opposite way it started, in silence and in friendship.

Your Boyfriend seems to be a musing on relationships, friendships, loneliness and perception. All of the characters are highly self-aware and articulate, with brief moments of amazing vocabulary and touching in their moments of honesty and openness. A phrase and idea that repeats itself is ?missing people?. The characters feel as though they are missing people in their lives, actual people or the idea of someone, a boyfriend, a girlfriend, true friends, so they use substitutes and stand-ins, which is a very human thing to do, everybody in the play does it, but Marci is the only one accused of doing it blatantly. While we never confirm her boyfriend is real, her boyfriend is real to her.

The idea of character perception is very fascinating to me, especially this week. By character perception I mean the idea putting someone or a character in a box you think they belong to, i.e. she?s the ?mean girl?, she?s ?the outsider?, he?s ?the friendly one, he?s ?the hunky one?.? Our media and entertainment perpetuate these ideas, and this week has been a fascinating conversation with the opinion piece by Polly Carl of HowlRound.com relating to the Guthrie season debate, and the recent release of (Spoiler Alert) Cabin in the Woods? which takes the horror movie character archetypes and tilts them on their heads. Your Boyfriend challenges the notion of character archetypes by pushing their boundaries and allowing them to do something wholly out of character and unexpected, something human.?

In an impromptu mini-interview with Larry, he had this to say about the characters, and specifically the words he chose to gift them with

??I try to throw in a word or phrase for all my characters in all my plays that are designed to throw an audience. In real life, language transcends class and race and gender and educational background, so I try to make all my characters, regardless of their backgrounds, hyper-articulate, but also, at the same time, grasping for meaning with words and in connection with others.?

(Full disclosure line, I have worked with and share amicable relationships and friendships with Larry Kurnofsky, Kirsten Hopkins, Debargo Sanyal and Risa Sarachen)
